Description:
Directed mutagenesis and chloroplast transformation of rbcL-W66Amber mt+ were used to create a C172S substitution (TGT-TCT) to investigate stress-induced degradation of Rubisco. The x-ray crystal structure of the mutant protein has been determined. This is the original mutant strain. It has been maintained with acetate medium in darkness to prevent selection for secondary mutations that may improve Rubisco stability.
