Description:
Directed mutagenesis and chloroplast transformation of rbcL-W66Amber mt+ were used to create an M42V substitution (ATG-GTA) which causes a small decrease in Rubisco carboxylase activity. This is the original mutant strain. It was created to investigate phylogenetic differences near large-subunit residue Gly-54 (see rbcL-G54D). The strain has been maintained with acetate medium in darkness to prevent selection for secondary mutations that may improve Rubisco function.
