Description:
Directed mutagenesis and chloroplast transformation of rbcL-W66Amber mt+ were used to create a Y226L substitution (TAC-TTA) in the Rubisco large subunit, which causes decreases in Rubisco carboxylation and holoenzyme stability. This mutant strain was created to investigate Rubisco degradation. Other nearby Tyr residues have been studied. The strain has been maintained with acetate medium in darkness to prevent selection for secondary mutations that may improve Rubisco function or stability.
