Our Methods for Target Bisulfite Sequencing of Bombyx Mori

Samples were subjected to targeted bisulfite sequencing followed by independent validation of selected sample differentially methylated regions using target gene DNA methylation sequencing. Using the methylation-specific multiplex PCR primer design software, the target gene after sulfite conversion is multiplex amplified, and the ultra-high-depth (above 1000X) methylation detection is carried out after the library is established.

Total DNA >50 ng, no significant degradation of DNA or protein contamination, as judged by gel electrophoresis on of DNA, gel electrophoresis test gel chart required.
Number of freshly cultured cells >106
Whole blood >0.5 mL, whole blood samples should be used in normal EDTA anticoagulation tubes, avoid using heparin anticoagulation tubes; cell samples should be live cell frozen samples
