Microscale thermophoresis Advantages: the amount of sample is small; there is no need to use antibodies, and there is no need to obtain high-purity proteins; not only can the interaction relationship between two biomolecules be qualitatively determined, but also the binding affinity can be quantitatively judged.
Disadvantages: no kinetic information and concentration analysis; if the interacting protein forms a complex with other molecules or competes with other molecules, the binding data will be affected; if the protein has post-translational modification, the data detected by different systems will drift; if the protein to be tested is not stable in the experimental system, which will affect the stability of the data.
Biacore Advantages: no labeling; high sensitivity; rapid detection; real-time quantitative testing.
Disadvantages: high cost.
