Liposomes have the following characteristics: (1) targeting; there are four types of chemical targeting, physical targeting, antibody targeting, and receptor targeting; (2) non-toxic safety; lecithin has a high safety level even when used in large amounts; (3) lipid-soluble and water-soluble compatibility: the phospholipid bilayer in the liposome wall material can carry lipid-soluble molecules while the vesicles can carry water-soluble molecules inside. Therefore, when wrapping small molecules, the efficacy of small molecules can be fully utilized.

Background

MicroRNAs are cellular RNA species that block the production of specific proteins by binding to and targeting mRNA for destruction. Due to their small size (i.e., 21-22 base pairs), microRNA mimics in mature double-stranded form can be transferred into the target cell. Liposomes can encapsulate polar or non-polar molecules. Their application can be extended by designing very small liposomes (<100 nm). The physicochemical and biophysical properties of liposomes can be easily manipulated for use in various fields. They can be fine-tuned to provide a rapid or delayed release of the encapsulated molecules.

  • Preparation of liposome-encapsulated microRNA

Liposome Encapsulated microRNA Services

The liposomal material is suspended in ethanol at various concentrations. This solution is then transferred to ethanol, and this stock solution is diluted to prepare a working solution. Once the desired dilution is reached, microRNA is added and incubated at room temperature. The solution is then placed in a separate vial containing PBS, magnetically stirred, and the working solution containing the microRNA is drawn into a glass syringe. Finally, the liposome suspension containing microRNA was dialyzed overnight.
