The encapsulation of DNA plasmids in liposomes has been achieved by various techniques with varying degrees of success, protecting DNA from nuclease attack and increasing its biological stability and bioavailability. Reverse-phase evaporation produces large monolayers of liposomes (average diameter of 400-500 nm), of which up to 40% can be captured using negatively charged phosphatidylserine (PS) containing vesicles. The dehydration-rehydration technique was adjusted to retain more than 60% of the plasmids in the oleic acid-containing vesicles. Still, most of the retained plasmids were released by squeezing through 0.45 μm filters, indicating very large vesicles. Smaller vesicles prepared by detergent dialysis with oleic acid re-added to the lipid mixture had a 15% capture rate in 200 nm liposomes.
