Liposome Encapsulated siRNA Services

Liposomes are composite structures made of phospholipids that are artificially created to encapsulate various types of nucleic acids within the aqueous core of liposomes. Due to the hydrophobic nature of the liposome outer membrane, it can be fused to other lipid bilayers, such as cell membranes, and deliver its contents via cell fusion. Lifeasible offers encapsulation of siRNA molecules into several standard liposome formulations (PC: cholesterol and DSPC: cationic lipids: polyethylene glycolated lipids: cholesterol) or custom liposome formulations. Custom experimental designs of liposomes can be used to provide siRNA protection (in the liposome core) in PEG-modified liposomes, where biodegradable lipids and low-toxicity polymers are used in the preparation process.

Background

siRNAs have sequence-specific targeting and their potential to inhibit protein translation. Liposomes composed of amphiphilic lipids, including phospholipids, can trap hydrophilic or hydrophobic materials in hydrophilic internal or hydrophobic lipid bilayers, respectively. Based on these properties, liposomes have been developed as delivery cargoes for various molecules, such as small molecules, nucleic acids (including siRNA and proteins), etc. Substances encapsulated in liposomes offer many advantages, such as preventing degradation and reducing substance toxicity.

Liposomes can be prepared by ultrasonication, liposome extrusion, and micro-fluidization methods, and liposome sizes can be customized from 50 nm to 400 nm.
