About Liposome Lamellarity

Among structural properties, lamellarity, the number of concentric lipid bilayers in a liposome, is one of the most relevant properties. Laminarity determines the stability of liposomes, the number of molecules that can be encapsulated, the release kinetics, and the properties of liposomes when interacting with cells. In addition, it determines liposomes' overall mechanical and dielectric properties, which are related to their interaction with cells.

Generally, the lamellarity can be determined using techniques such as NMR, X-ray small angle scattering, and fluorescence spectroscopy. These techniques provide an average lamellarity of the liposome population. Laminarities can also be determined at the level of individual liposomes by considering imaging techniques such as cryo-electron microscopy, cryo-fracture electron microscopy, and light microscopy or mechanical techniques such as micropipette aspiration. Recently, atomic force microscopy (AFM) and force spectroscopy measurements have also been developed.
