Background

Liposomes are spherical artificial vesicles made from cholesterol and natural non-toxic phospholipids. The properties of liposomes vary greatly depending on lipid composition, surface charge, size, and preparation method. The choice of liposome bilayer components determines the "rigidity" or "fluidity" and the charge of the bilayer. Liposomes are generally available in particle sizes ranging from 30 nm to several microns. They consist of one or more lipid bilayers around aqueous units, where the polar head group is located in the internal and external channels of the aqueous phase. On the other hand, the self-aggregation of polar lipids is not limited to conventional bilayer structures that depend on the molecular shape, temperature, environment, and preparation conditions but can self-assemble into various types of colloidal particles. The encapsulation technique of liposomes can be used in agriculture by wrapping different types of molecules for application in specific tissues. Liposomes can trap hydrophobic and hydrophilic compounds, avoid the decomposition of entrapped assemblies, and release entrapped material at the specified target.

Liposome Preparation Methods

  • Ultrasonication method

Ultrasonication is probably the most used method for lipid dispersion, especially for the fabrication of small unilamellar vesicles (SUVs). In this method, liposomes are passively produced using either a bath-type ultrasonicator or a probe-type ultrasonicator.

  • Extrusion method

The extrusion method uses a squeezing process and pushes multilayer vesicles through small pores to disperse the lipids.

  • Freeze-thaw liposomes method

In this method, the SUVs are frozen for a short period and then thawed over a long period. This disperses the lipids and leads to the formation of large unilamellar vesicles (LUVs).

  • Other methods

Several other dispersion methods, including lipid membrane hydration, micro-emulsification, membrane extrusion, and drying reconstituted vesicles.
