The rapid determination of pollen viability by the color reaction of Pollen Viability Test Kit (Peroxidase Assay) allows experimental prediction and assessment of pollen fertility to aid breeding.

Testing Mechanism
Viable pollen contains an active peroxidase enzyme. Peroxidase can use hydrogen peroxide to oxidize various polyphenols and aromatic amines to produce color, and the activity of pollen can be assessed based on the color change.
Kit Components
1. Reagent (A)
A1: Aniline staining solution (10 mL)
A2: Naphthol staining solution (10 mL)
A3: Aromatic buffer solution (10 mL)
Before use, Reagent (A) needs to be mixed in the ratio A1:A2:A3=1:1:1.
2. Reagent (B) Oxidizer (2*1 mL)

Vigor Judgment Criteria
1. Purplish red color means strong vitality.
2. Light red means weak vigor.
3. Colorless means no vitality or sterility.
Pollen Vigor Calculation Formula
Observe and count 100 pollen grains to calculate the percentage of viable pollen. The calculation formula is as follows:
Percent pollen viability (%) = Number of viable pollen / 100 × 100%
Notes
1. The oxidizer is corrosive, so handle carefully to avoid the oxidizer volatilization, otherwise the staining power will be reduced.
2. The pollen needs to be completely submerged in the staining solution when staining.
3. For your safety and health, please wear lab coat and disposable gloves to operate.
