Synechocystis sp. PCC 6803 is unicellular, freshwater cyanobacteria, with the capacity of both phototrophic growth by oxygenic photosynthesis during light periods and heterotrophic growth by glycolysis and oxidative phosphorylation during dark periods. And it is naturally transformable, with the genome size of 3.6 Mb, containing a circular chromosome and 7 plasmids size ranging from 2.3 to 120 kb.
Many genetic tools can be applied in PCC 6803, which makes it an ideal platform of biosynthesis. For example, genes involved in RuBisCO and photosystem have been promoted for ethanol producing, genes regulating responding to environment have been modified to generated strains capable of adapting stressful environment. By overexpressing stress-resistant proteins can improve its tolerance to the toxic products. Chemicals like acetone, ethylene and isoprene can also be produced by introducing related pathways into PCC 6803. What’s more, biopolymer like polyesters can also be produced by PCC 6803.
