  • Agrobacterium-mediated method. Integration of exogenous genes is achieved by using Agrobacterium to carry plasmids containing the target genes and infecting the healing tissues or young tissues of Cucurbita pepo through wounding. As a natural plant gene transformation system, Agrobacterium-mediated transformation is a purely biological process with obvious advantages over other transformation methods, mainly including high transformation frequency, good expression, and stable inheritance.
  • Viral vector method. Viral vectors based on tobacco ringspot virus (TRSV), carrying specific gene fragments or miRNAs, cause different phenotypes after infiltration of Cucurbita pepo, and are used to study gene functions. TRSV-based vectors were developed to carry gene fragments or miRNA specific to melon crops and infest Cucurbita pepo.
