  • Optimization of Agrobacterium method for transforming Eremochloa ophiuroides using GUS transient expression We performed GUS staining of transformed healing tissues to detect transient expression of genes. The optimized Agrobacterium-mediated transgenic method for Eremochloa ophiuroides was obtained by designing different treatments using healing tissues with varying times of incubation, hypertonic medium, AS pre-culture, and ultrasound-assisted infiltration using GUS staining for transient expression.
  • Transfection of target genes into the Eremochloa ophiuroides genome using the gene gun method. We used mature embryonic healing tissues of Eremochloa ophiuroides as the recipient for transformation by the gene gun method, and the healing tissues obtained from the transformation were cultured to obtain regenerated plants, which could be screened by PCR to see if positive plants were obtained.
