Lilium has not only good ornamental value but also edible and medicinal functions. Due to the lack of a stable and efficient genetic transformation technology system, the research on the gene function of Lilium and germplasm innovation through molecular means has been greatly restricted for a long time. So far, about dozens of multiple methods have been used to establish genetic transformation receptor systems for Lilium, a perennial bulbous plant. With the development of molecular biology, the ability to produce new Lilium varieties with stress resistance, and disease and good quality through transgenic methods is of great significance for Lilium breeding.

What do we offer?

We established a genetic transformation system for Lilium using the Agrobacterium rhizogenes-mediated genetic transformation method. Concentrations of screening agents suitable for Lilium transformation, strain selection, determination of the concentration of the bacterial solution, infiltration time, and other transformation conditions were determined to achieve the highest transformation efficiency for Lilium.

Currently, the main methods we applied to Lilium gene transformation are the Agrobacterium-mediated method, gene gun method, and electro-excitation method. Agrobacterium-mediated method is the most widely used in Lilium genetic transformation. While the gene gun method is a new method of genetic transformation emerging in recent years, the research of the gene gun method in Lilium genetic transformation is mainly to study the expression of marker genes in the recipient system. In addition, the electrostimulation method utilizes the pulses generated by high-voltage discharges to create reversible "micropores" in the cell membrane, thus allowing exogenous DNA and other macromolecules to enter the cell through this channel.

Establishment of an efficient regeneration system and genetic transformation of Lilium. We have established an efficient regeneration system for Lilium using its leaf tissue as an explant. It was found that the differentiation ability of Lilium leaf tissues was stronger than that of petiole tissues. We used the Agrobacterium rhizogenes-mediated method to transform Lilium with a target gene and obtained a batch of thaumatin-resistant plants through screening and differentiation induction. Finally, positive plants were obtained using PCR assay. This technical support service we provided provides a research basis for carrying out Agrobacterium-mediated genetic transformation of Lilium on a large scale and improving the quality of Lilium.

Our research objectives

  • Improvement of ornamental traits. Lilium has good ornamental plants, and genetic transformation can be used to improve its aesthetic traits. It changes traits such as flower color, shape, size, fragrance, and longevity.
  • Resistance to pests and diseases. Lilium is susceptible to various pests and diseases. Transgenic approaches can enhance resistance to fungal, viral, or bacterial pathogens. This may involve the introduction of genes encoding anti-microbial proteins, enzymes that detoxify harmful compounds, or genes that confer resistance to specific pathogens.
  • Production of pharmaceutical compounds. Some Lilium species contain bioactive compounds with potential pharmaceutical uses. Genetic transformation could be used to increase the production of these compounds or to introduce new biosynthetic pathways into Lilium plants.

In various improvement studies based on Lilium genetic transformation, we not only detect the integration of exogenous genes but also observe the stable, normal, and high-level expression of exogenous genes.
