Rice anther culture is the process of using aseptic operation techniques to inoculate rice anthers that have developed to a certain period onto an artificial medium, inducing dedifferentiation to form healing tissues by altering the developmental program of pollen grains and then allowing the healing tissues to re-differentiate to form haploid plants. Haploid plants undergo colchicine or natural doubling to become complete diploid plants. Combining anther culture with other breeding techniques can rapidly purify breeding materials, effectively shorten the breeding cycle, improve selection efficiency, and accelerate the breeding process.
Conventional rice breeding methods need to be separated for many generations to form stable varieties. At the same time, anther culture technology has the advantages of shortening the breeding period and rapidly stabilizing breeding materials. The use of anther culture technology as a breeding aid is highly effective in the creation of original materials, selection of new varieties, and purification of sterile lines. We used rice anthers as materials to obtain independent healing tissues quickly and with high quality for further differentiation and regeneration into plants. When rice is in the pregnant spike stage and the leaf cushion spacing is 5-15cm (this period is the mononuclear leaning stage of pollen differentiation), cut the spike, peel off the leaf sheaths, cut the young spike into a length of 5cm, peel off the anthers with pointed forceps, inoculate the anthers into the medium, and when the healing tissues are grown out, transfer them to the differentiation medium, and differentiate the green seedling.
