The shuttle vector is a vector that can be replicated, proliferated, and selected in two different types of host cells, such as plasmids that can be replicated and expressed in bacteria, mammalian cells, yeast, and other different hosts. Lifeasible can construct various shuttle plasmid vectors containing target genes according to the customer's needs, incorporating different promoters, screening tags, resistance, and so on. These plasmids can proliferate in prokaryotic and eukaryotic cells. Typical examples are adenovirus shuttle plasmids and yeast shuttle plasmids, which can replicate in eukaryotic cells (e.g., mammalian cells, yeast) and proliferate in prokaryotic cells (e.g., E. coli).

Types that can be constructed

  • In mammalian / E. coli shuttle vector

Can be expressed in mammalian cells and also has replication elements of E. coli, such as eukaryotic expression vectors.

  • E. coli/ Gram-positive bacterial shuttle vectors

Can transfer the target gene to Bacillus or Coccidioides.

  • E. coli/ Yeast shuttle vectors

In addition to playing an essential role in the study of the cell biology of eukaryotes, yeast has also shown great power in serving as a eukaryotic expression system for proteins. We constructed an E. coli / Saccharomyces cerevisiae shuttle plasmid vector containing replication start and selection markers from E. coli and Saccharomyces cerevisiae, respectively, and an additional polyclonal site region. This vector type is popular in genetics research because it can replicate in both E. coli and Saccharomyces cerevisiae cells. It allows you to freely transfer genes back and forth between two different host cells and to have the ability to study the expression activity and other regulatory functions of the target gene in both host cells singly or simultaneously.

  • Other shuttle vectors

Expression vectors or other vectors are used in plants, insects, and other cells. They generally have elements that replicate in E. coli and, therefore, have the characteristics of shuttle vectors.

Building methods

  • The customer provides the plasmid backbone and gene template; the customer needs to ensure that the vector and gene template provided are correct.
  • Customers provide gene templates but no plasmid backbone; customers need to specify their experimental needs, and we will help you determine the best plasmid backbone.
  • If you do not have a gene template or plasmid backbone, you need to provide clear gene information and construction requirements to ensure that the plasmid constructed by us can achieve your final experimental purpose.
