Inter Simple Sequence Repeats Technology Service

Inter-simple Sequence Repeat (ISSR) is a new molecular marker technique developed based on SSR. Its basic principle is to add 2-4 random nucleotides to the 3' or 5' terminal of SSR sequence, amplify a sequence with the reverse arrangement of SSR on both sides by PCR, and then use electrophoresis technology to analyze its polymorphism according to the presence or absence of bands and their relative position.

ISSR markers are based on the widespread presence of SSRs in plants, and primers are designed using the SSRs themselves, which are commonly found in plant genomes, without the need for pre-cloning and sequencing. The primers used for ISSR-PCR amplification are usually 16 to 18 base sequences, consisting of tandem repeats of 1 to 4 bases and several non-repetitive anchor bases, thus ensuring that the primers bind to the 5' or 3' terminal of the SSRs in genomic DNA, resulting in repeats located in reverse alignment and not too widely spaced between of genomic segments for PCR amplification.
