Volvariella volvacea Transformation

Volvariella volvacea is a saprophytic fungus native to tropical and subtropical regions with high temperatures and rainfall and is mostly grown on fiber-rich substrates. Volvariella volvacea are rich in many essential amino acids such as serine, alanine, and glycine. It also has high medicinal value and contains many vitamins, which can improve human health and reduce the absorption of cholesterol. Moreover, Volvariella volvacea can secrete cellulase, hemicellulase, ligninase, and other decomposition enzymes, and can grow on a variety of substrates containing cellulose and lignin, so it is widely cultivated and has a short production cycle. It is a class of edible mushrooms with high economic value and broad development prospects. However, because of its intolerance to low-temperature freezing, its mycelium will die by autolysis under conventional low-temperature freezing conditions, so genetic transformation breeding of Volvariella volvacea has been a hot research topic.

We offer Agrobacterium strains such as EHA105 (RifR), EHA101 (KanR), LBA4404 (RifR), A281 (RifR, NalR), LBA4404, GV3101, etc. We screen for resistance with HygR. We also offer plasmids with pCAMBIA 1301, pBGgHg, pLg-afp235 and other series. The commonly used reporter genes are GFP, GUS, and DsRed2. We also screen the AS content in advance for optimal concentration during pre-culture and co-culture. The reason for doing this is to match the optimal AS content for each experiment, and we also strictly control the co-culture temperature and time, the selection of Agrobacterium strains, and other influencing factors to ensure high transformation efficiency.
