Lolium perenne is an important pasture grass, as well as an important lawn grass. It was originally distributed in Asia, Europe, North Atlantic islands, and temperate regions of North Africa, and it is now planted all over the world. Lolium perenne has a strong tolerance to trampling and cold but is less tolerant to drought and heat. The cultivation of new varieties of Lolium perenne has a very significant effect on improving forage yield and quality, improving resistance to adverse environments, improving natural grassland, establishing artificial grassland, maintaining soil and water, preventing wind and sand, and improving the ecological environment of human beings, etc. Therefore, it is of great significance to explore the use of biotechnology to genetically transform Lolium perenne to obtain high-quality Lolium perenne varieties with excellent agronomic traits.
Lifeasible established a genetic transformation system of Lolium perenne grass gene gun to lay a good foundation for its transgenic operation, and introduced exogenous genes into Lolium perenne to improve its survival ability in adverse environments, such as drought, low temperature, and disease. Establishing this transformation system lays the foundation for directed molecular design to improve Lolium perenne varieties. In addition, we are constantly searching for a more efficient genetic transformation system, as well as exploring ways to simplify and optimize the transformation conditions and screening sensitive varieties, to obtain higher transformation efficiency and stably expressed transgenic plants and laying the foundation for the transfer of target genes, such as drought-resistant genes, and the acquisition of drought-resistant new materials.
We generally transform the healing tissues of Lolium perenne using Agrobacterium tumefaciens, where Agrobacterium tumefaciens contains a plasmid carrying target and selective resistance genes, and the said healing tissues are induced from mature embryos of Lolium perenne. We induced the healing tissues by removing the mature embryos of Lolium perenne on the induction medium. The induced healing tissue was co-cultured with Agrobacterium rhizogenes to transform the healing tissue. Freshly induced healing tissue can be used, or healing tissue from a successor can be used. If a succeeding healing tissue is used, the healing tissue should preferably have been succeeded for no more than half a year. After completing the transformation step, we will also perform recovery decontamination culture on the transformed healing tissues. We have used healing tissues induced from mature embryos of perennial Lolium perenne, infested with Agrobacterium with antibiotic resistance genes, and screened for antibiotics to obtain resistant plants. Our transformed plants will be verified by PCR, GUS tissue staining and Southern blotting, aiming to provide stably expressed transgenic Lolium perenne plants.
