Optical Immunosensors for Plant Pathogen Detection

Optical immunosensors use a photosensitive element as an information transducer, work on optical principles, and are an essential member of the immunosensor family. The photosensitive elements are optical fibers, waveguide materials, gratings, etc. Biorecognition molecules are solidified on the sensor, and by interacting with the optics, a changing optical signal is generated, and the immune response is detected by detecting the changing optical signal. Immunofluorescence method

This method combines immunological methods with fluorescent labeling techniques to localize antigens in tissues or cells using antigen-antibody reactions. Generating fluorescence in an assay system is not complicated; one way is to label the fluorescent groups (TAMRA, FAM, Cyc3). Another way is to add embedded-type fluorescent dyes to track the formation of DNA double strands in the reaction. We use calcein added to the loop-mediated isothermal amplification (LAMP) system, which can be used for qualitative judgment by the naked eye or precise quantification with the aid of an instrument.

  • We can provide microsphere immunoassay (MIA)-based multi-detection systems for simultaneously detecting four plant pathogens, including Acidovorax avenae subsp. Citrulli (AAC), Chilli vein-banding mottle virus (CVbMV), Watermelon silver mottle virus (WSMoV), and Melon yellow spot virus (MYSV). The sensitivity of the assay was much higher than that of enzyme-linked immunosorbent assay (ELISA).

Surface plasmon resonance (SPR)

SPR is capable of instantaneous measurement of the sample to be tested and can monitor dynamically so that we can measure the dissociation constant (Kd) and binding constant (Ka) between protein-protein or protein-DNA by SPR.

  • We constructed a label-free SPR immunosensing system using gold nanoparticles (AuNRs), which can be used to detect both Cymbidium mosaic virus (CymMV) and Odontoglossum ringspot virus (ORSV). We use AuNRs as a detection layer to modify virus-specific antibodies to have a broader spectral range, which helps to reduce the problem of color interference caused by the sample matrix.
  • Our system can also be used to detect Tobacco mosaic virus (TMV).
